Synergistic Antibacterial Effect of Lup-20 (29)-ene-3α, 23-diol from Glochidion daltonii (MÜII. Arg.) Kurz. and Antibiotics on Opportunistic Bacteria

Abstract

The objectives of this research were to tested antibacterial activity of Lup-20 (29)-ene-3α, 23-diol, isolated from hexane extract from the stem of Glochidion daltonii (MÜII. Arg.) Kurz. against some opportunistic bacteria.  The screening test of antibacterial activity were shown that Lup-20 (29)-ene-3α, 23-diol could inhibit the growth of E. coli ATCC 25922, P. aeruginosa ATCC 27853 and S. marcescens by MICs of 128-512 µM.  Also, synergistic effect and time kill assay was tested using Lup-20 (29)-ene-3α, 23-diol mixed with antibiotics against bacteria.  The result were shown that Lup-20 (29)-ene-3α, 23-diol mixed with tetracycline indicated the best synergistic effect against P. aeruginosa  ATCC 27853 with the FICI of 0.28.  Moreover, the results indicated the indifferent effects against E. coli ATCC 25922 when using Lup-20 (29)-ene-3α, 23-diol,  mixed with ampicillin or tetracycline and the result indicated equal FICIs as 1.007.  For finding the best the time of Lup-20(29)-ene-3α, 23-diol mixed with tetracycline to kill P. aeruginosa ATCC 27853, it was indicated that concentration, mostly dccreased the number of P. aeruginosa ATCC 27853 was using 1/16 MIC of MIC alone of tetracycline and 1/16 MIC of MIC alone of Lup-20 (29)-ene-3α, 23-diol.  Also, the best times, mostly decreased the growth of against P. aeruginosa ATCC 27853, were 8th hour.  The result obtained in this research indicated that Lup-20 (29)-ene-3α, 23-diol had an effective synergistic effect when mixed with tetracycline,  and indicated synergistic effect against some opportunistic bacteria.
